Tangential Flow Filtration (TFF), also called Cross Flow Filtration (CFF), is a rapid and efficient method for filtration and separation of bioprocessing fluids. In tangential flow system configuration, the particels or containments thar are too large to pass through the membrane pores are retained on the upstream side and are continuously swept away from the surface by the flow stream across the surface. Unlike normal flow filtration (NFF), TFF feature reduces the polarized surface layer build up on the filter membrane which can gradually decrease the flow rate and eventually lead to an unacceptable pressure differential. It can also be operated continuously at relative high loads without fouling. This makes TFF ideal for fine size based separation, such as ultrafiltration/diafiltration (UF/DF) application. TFF is commonly used in separation of proteins from buffer components for buffer exchange, desalting, or concentration prior to final product fill and finish. Important process parameters include retentate flow rate, tranmembrane differential pressure, temperature, and product properties such as concentration, viscosity and contents.
